The Nanotemper Prometheus is used to test protein stability and protein aggregation by either chemical or thermal denaturation.
The instrument monitors the intrinsic fluorescence signal of a protein sample as a measure of its folding state. A ratiometric measurement of the fluorescent signal is plotted against either increasing concentration of a chemical denaturant or increasing temperature, to determine the Tm of a protein.
The instrument can test 1 to 48 samples at a time, using as little as 10 uL in volume and 5ug/mL in concentration.
The sample is loaded into a capillary, placed into the instrument and a program is started. The results are available within minutes. The method may be useful for small or large proteins, enzymes,antibodies and membrane proteins and is particularly good at screen buffers and for testing storage conditions.
